The Morphology of Bull Spermatozoa

چکیده

It has been known since the nineteenth century that the microscopical structure of living cells may be modified by fixation and by post-mortem or sub-mortem changes (Hardy, 1899), but inadequate attention seems to have been paid to factors which may injure spermatozoa before fixation and microscopical examination. One such factor is the effect of sudden cooling. If semen is exposed to a sudden fall in temperature there is an irreversible decline in the activity of the spermatozoa (Chang & Walton, 1940), and this reduction in activity is associated with an increase in the number of dead, i.e. eosinophil cells (Lasley, Easley & McKenzie, 1942). This phenomenon of 'temperature shock' is encountered in bull semen cooled abruptly by pipetting semen at 30 C. into an aqueous solution of nigrosin and eosin at 20 C. (Hancock, 1951). The depressed rate of respiration and fructolysis of temperature-shocked samples can be correlated with the increase in the proportion of eosinophil cells (Bishop & Hancock,

Effect of the Addition of Insulin Like Growth Factor-I (IGF-I) on Cryopreservation of Hariana Bull Semen

This experiment was designed to study the effect of insulin like growth factor-I (IGF-I) on cryopreservation of Hariana bull semen. For this purpose, semen ejaculates were collected from four Hariana bulls using artificial vagina at biweekly interval. The semen samples which possess more than 70% initial progressive motility and above 500 million/mL spermatozoa concentration was subsequently su...

Spermatozoa Molecules in Relation to Bulls Fertility

Bull fertility may be defined as the process by which spermatozoa fertilize and activate the ovum and then support embryonic development. Bull fertility is a complex trait having relatively low heritability and plays a vital role for efficient production and reproduction of bovine. Various mechanisms involved in regulating bull fertility associated phenotype and reliable biomarkers are poorly d...

Bull Fertility and Its Relation with Density Gradient Selected Sperm

Background sperm selection is generally used to describe methods for separation of spermatozoa for in-vitro-assisted reproduction. Few studies reported relation in vivo fertility and semen parameters after selection of fertile spermatozoa; hence, our research attempt to assess different semen parameters after post-thaw (PT) or after selecting sperm sample (SSp), using density gradient separatio...

O-12: Studies on Sequestration of PDC-109 Protein on Cryodamage and In Vitro Fertility of Crossbred Bull Spermatozoa

Background: Plasma membrane of the spermatozoa interacts with and is altered by abundant seminal PDC-109 present in their immediate milieu in vivo with damaging effect in a time- and concentration- dependent manner. Therefore we hypothesized that sequestration of detrimental protein from ejaculates would be beneficial for cryopreservation of sperm cells. To this aim we evaluated the effect of s...

The effect of sperm concentration in the ejaculate on morphological traits of bull spermatozoa.

Experiments were performed on 75 ejaculates obtained from 19 bulls representing different cattle breeds used at the Masovian Centre for Animal Breeding and Reproduction in Łowicz. Fresh ejaculates were measured in respect to their volume and sperm count in the ejaculates was determined. The ejaculates were classified based on the criterion of sperm concentration and divided into five groups. Sp...
